I’ve used 4-5 different techniques to look at the heart. But I’ve done each of those experiments/techniques many many times. Science is all about making sure what you found is truly there, so you have to repeat some experiments over 3 times each to make sure that your results are real.
The standard experiment I do, is putting liquid into the lungs of rats and measure the volume of liquid before and after different chemicals are added. These chemicals either activates or blocks enzymes and channels. I’ve also done similar experiments on sheep. Away from my work on animals, I’ve grown cell and used antibodies to identify whether the cells had the channels I am interested in.
